Hoechst 33342 molecular probes
Nettet(Molecular Probes: C-1430). Hoechst 33342, a bisbenzimide dye, is a cell membrane permeant, minor groove-binding DNA stain that fluoresce bright blue upon binding to DNA. The dye is quite soluble in water and relatively non-toxic (Molecular Probes: H-1399, H-3570). Hoechst 33342 stains both, live and dead cells. Material and Methods Cell Culture Nettet1. jan. 2009 · Hoechst 33342 is the fluorophore used routinely to measure DNA in X- and Y-chromosome-bearing mammalian sperm so they can be separated by flow sorting. A difference of <3% in DNA mass can be detected. This synthetic dye consists of two adjacent benzimidazole rings with N-methyl-piperazine and phenolic groups at the ends.

NettetHoechst 33342 and 33258 are quenched by bromodeoxyuridine (BrdU), which is commonly used to detect dividing cells. Hoechst 33342 exhibits a 10 fold greater cell-permeability than H 33258. Cells can integrate BrdU in newly synthesized DNA as a substitute for thymidine. NettetAfter removing MitoTracker Red CMXRos, cells were treated with 200 nM MitoTracker Green FM (Molecular Probes) and incubated at 37 °C for 30 min. After removing MitoTracker Green FM, cells were treated with Hoechst 33342 (Dojindo) and incubated at 37 °C for 30 min. The fluorescence intensity of MitoTracker Red CMXRos, ...
